Adansonia digitata L. (Bombacaceae) is a native deciduous tree of African savannas and is known for its unique medicinal properties and charismatic attractive appearance. The plant has limited distribution in India; it is not found universally and is facing the risk of getting extinct.Present study was carried out to establish a standardized protocol for in vitro bud break using tissue culture approaches. Effect of basal media and auxins/cytokinins was investigated on bud break. Results indicate that MS medium induced bud break in 91.6% explants followed by Gamborgs (B5) (86.6%), Woody Plant Medium (WPM) (83.3%), LP (61.6%) and Nitsch and Nitsch (NN) (55.0%) respectively.Bud break in nodal segments could be increased to 98% on Murashige &Skoog (MS) basal medium (full strength) with Benzyl amino purine (BAP) at concentrations (0.4 and 0.6 mg/l).BAP + kinetin induced higher number of multiple shoots per explant. The maximum average shoot length (3.63cm) was observed on medium with BAP (0.6mg/l) + kinetin (2.0mg/l). The conservation strategy under Indian sub-tropical states will help in studying its medicinal properties for further research on this multipurpose tree species.
